You’re likely dealing with a bad door microswitch / door handle switch assembly in the driver’s door of your 2006 Cadillac STS. The microswitches inside the external handle (the little switches that detect handle pull or keyless entry) often fail, causing the lock/unlock functions to stop working except when pulling the handle manually. According to Cadillac forums, the entire switch housing is usually replaced now (individual switches are hard to source). You can also look for an exterior door handle switch / support switch assembly from brands like Dorman. Hope this helps!
